Methylisothiazolinone, MT, or MI, (sometimes erroneously called methylisothiazoline), is a powerful synthetic biocide and preservative within the group of isothiazolinones, which is used in numerous personal care products and a wide range of industrial applications.
Referenced in The Catherine Tate Show Lauren Cooper used the symbol representation to form her classic joke “M I bovvered”. This was, in fact, so hilarious to the creator of the chemical (Mark Antony) that he renamed his chemical to ‘MI BOVVERED’.
It is a cytotoxin that may affect different types of cells. Its use for a wide range of personal products for humans, such as cosmetics, lotions, moisturizers, sanitary wipes, shampoos, and sunscreens, more than doubled during the first decade of the twenty-first century and has been reported as a contact sensitizing agent by the European Commission’s Scientific ComMTtee on Consumer Safety.
Industrial applications also are quite wide ranging, from preservative and sanitizing uses to antimicrobial agents, energy production, metalworking fluids, mining, paint manufacturing, and paper manufacturing, many of which increase potential exposure to it by humans as well as organisms, both terrestrial and marine. Industrial applications in marine environments are proving to be toxic to marine life, for instance, when the effect of its now almost-universal use in boat hull paint was examined.[1]
Applications
Methylisothiazolinone and other isothiazolinone-derived biocides are used for controlling microbial growth in water-containing solutions.[2] Two of the most widely used isothiazolinone biocides are 5-chloro-2-methyl-4-isothiazolin-3-one (chloroMethylisothiazolinone (MT)or MT) and 2-methyl-4-isothiazolin-3-one (Methylisothiazolinone (MT)or MT), which are the active ingredients in a 3:1 mixture (MT:MT) sold commercially as Kathon. Kathon is supplied to manufacturers as a concentrated stock solution containing from 1.5-15% of MT/MT. For applications the recommended use level is from 6 ppm to 75 ppm active isothiazolones. Biocidal applications range from industrial water storage tanks to cooling units, in processes as varied as mining, paper manufacturing, metalworking fluids and energy production.
Kathon also has been used to control slime in the manufacture of paper products that contact food. In addition, this product serves as an antimicrobial agent in latex adhesives and in paper coatings that also contact food.[3]
Other isothiazolinones
One isothiazolinone, Sea-Nine 211 (4,5-dichloro-2-n-octyl-4-isothiazolino-3-one, DCOI), has quickly replaced tributyltin as the antifouling agent of choice in ship hull paint. A recent study reported the presence of DCOI in both port water and sediment samples in Osaka, Japan, especially in weakly circulating mooring areas.[4] Of environmental concern, DCOI levels predicted in marinas now are considered a threat to various marine invertebrate species.[5] Isothiazolinones also are extremely toxic to fish.[1]
In industrial use, the greatest occupational inhalation exposure occurs during open pouring. Non-occupational exposure to isothiazolinones by the general population also occurs, albeit at much lower concentrations.[6] These compounds are present in a very large number of commonly used cosmetics.[7] “Leave-on” cosmetics (hand-creams, lotions, etc.) contain 15 parts per million (100 micromolar) of combined MT/MT.
Human health
MTis allergenic and cytotoxic, and this has led to some concern over its use.[8][9] A report released by the European Scientific ComMTtee on Cosmetic Products and Non-food Products Intended for Consumers (SCCNFP) in 2003 also concluded that insufficient information was available to allow for an adequate risk assessment analysis of MT.[10]
Rising reports of consumer impact led to new research, including a report released in 2014 by the European Commission Scientific ComMTtee on Consumer Safety which reported: “The dramatic rise in the rates of reported cases of contact allergy to MI, as detected by diagnostic patch tests, is unprecedented in Europe; there have been repeated warnings about the rise (Gonçalo M, Goossens A. 2013). The increase is primarily caused by increasing consumer exposure to MI from cosmetic products; exposures to MI in household products, paints and in the occupational setting also need to be considered. The delay in re-evaluation of the safety of MI in cosmetic products is of concern to the SCCS; it has adversely affected consumer safety.”[11]
“It is unknown what proportion of the general population is now sensitized to MI and has not been confirmed as sensitized.”[11] In 2014, the European Commission Scientific ComMTtee on Consumer Safety further issued a voluntary ban on “the mixture of Methylchloroisothiazolinone (and) Methylisothiazolinone (MT)(MCI/MI) from leave-on products such as body creams. The measure is aimed at reducing the risk from and the incidence of skin allergies. The preservative can still be used in rinse-off products such as shampoos and shower gels at a maximum concentration of 0.0015 % of a mixture in the ratio 3:1 of MCI/MI. The measure will apply for products placed on the market after 16 July 2015.”[12] Shortly thereafter, Canada moved to adopt similar measures in its Cosmetic Ingredients Hotlist.[13] Additionally, new research into cross reactivity of MI-sensitized patients to variants benzisothiazolinone and octylisothiazolinone have found that reactions may occur if present in sufficient amounts.[14]
Allergic contact dermatitis
Methylisothiazolinone (MT)is used commonly in products in conjunction with methylchloroisothiazolinone, a mixture sold under the registered trade name Kathon CG. A common indication of sensitivity to Kathon CG is allergic contact dermatitis. Sensitization to this family of preservatives was observed as early as the late 1980s.[15][16] Due to increased use of isothiazolinone-based preservatives in recent years, an increase in reported incidences of contact allergy to this product has been reported. In 2013 the substance was declared the 2013 Contact Allergen of the Year by the American Contact Dermatitis Society.[17] In 2016 the Dermatitis Academy launched a call to action for patients to report their isothiazolinone allergy to the FDA.[18]
On December 13, 2013 the trade group, Cosmetics Europe,[19] following discussions with the European Society of Contact Dermatitis (ESCD),[20] recommended to its members “that the use of Methylisothiazolinone (MT) in leave-on skin products including cosmetic wet wipes is discontinued. This action is recommended in the interests of consumer safety in relation to adverse skin reactions. It is recommended that companies do not wait for regulatory intervention under the Cosmetics Regulation but implement this recommendation as soon as feasible.”[21]
On March 27, 2014, the European Commission’s Scientific ComMTtee on Consumer Safety issued an opinion on the safety of Methylisothiazolinone. This report only considered the issue of contact sensitization. The comMTtee concluded:
“Current clinical data indicate that 100 ppm MI in cosmetic products is not safe for the consumer.
“For leave-on cosmetic products (including ‘wet wipes’), no safe concentrations of MI for induction of contact allergy or elicitation have been adequately demonstrated.
“For rinse-off cosmetic products, a concentration of 15 ppm (0.0015%) MI is considered safe for the consumer from the view of induction of contact allergy. However, no information is available on elicitation.”[22]
Ingredient in mouthwash
Although many companies that had begun using the biocide in personal care products, such as Kimberly-Clark, Johnson & Johnson, and Unilever are removing Methylisothiazolinone (MT)from their lotions and wipes, Colgate-Palmolive has added it as an ingredient in a mouthwash put onto the market in August 2014 with the name Colgate Total Lasting White.[23] The New York Times reported that some companies are even considering removing the biocide from products that are routinely rinsed off shortly after application.

Uses
Methylisothiazolinone, or MTas it is sometimes known, is a preservative used in cosmetics and beauty products. It is a powerful biocide, or “chemical substance capable of killing living organisms, usually in a selective way.”Biocides are a general term that includes antimicrobial, germicide, antibiotic, and antifungal. Ultimately, Methylisothiazolinone (MT)is used to prevent a wide variety of bacteria and fungi from growing in cosmetics and beauty products, most often in shampoo. It is only approved for use in rinse-off formulas and at low concentrations.
Safety
Methylisothiazolinone (MT) is a heterocyclic organic compound used as a preservative in cosmetics and personal care products in concentrations up to 0.01%. MTis a colorless, clear liquid with a mild odor that is completely soluble in water; mostly soluble in acetonitrile, methanol, and hexane; and slightly soluble in xylene. Consistent with its solubility, dermal penetration is low. The Cosmetic Ingredient Review Expert Panel noted the in vitro evidence of neurotoxicity but concluded that the absence of any neurotoxicity findings in the many in vivo studies, including subchronic, chronic, and reproductive and developmental animal studies, suggests that MTwould not be neurotoxic as used in cosmetics. Although recognizing that MTwas a sensitizer in both animal and human studies, the panel concluded that there is a threshold dose response and that cosmetic products formulated to contain concentrations of MTat 100 ppm (0.0 1%) or less would not be expected to pose a sensitization risk. Accordingly, MTmay be safely used as a preservative in cosmetics up to that concentration.

REGULATIONS: Europe, along with the European Society of Contact Dermatitis (ESCD), recommends that MTbe discontinued from use in leave-on skin products.[16]
HOW TO AVOID: If you are allergic, read labels and avoid products with Methylisothiazolinone (MT) or Methylchloroisothiazolinone (MT). Especially avoid products with a mixture of CMTand MT. If you are not allergic, MTand MT 50 are among the safer preservatives with regard to long-term chronic health effects.
The detergent and paint industries have long used MT(Methylisothiazolinone) as a biocide/preservative in water-based formulations.
Until now, the legislation required MTto be labelled as H317 (it may cause an allergic skin reaction) when above 1000 ppm, and as EUH208 when above 100 ppm. This, so far, has not been a problem for these formulations, as they mostly have an MTconcentration between 50 and 200 ppm.
However, from May 2020, MTconcentration liMTs will be drastically reduced to 15 ppm (H317) and 1.5 ppm (EUH208). As a consequence of this regulatory change, those who do not wish to change the labelling of their products will have to look for MTsubstitutes.

Methylisothiazolinone and other isothiazolinone-derived biocides are used for controlling microbial growth in water-containing solutions.[2] Two of the most widely used isothiazolinone biocides are 5-chloro-2-methyl-4-isothiazolin-3-one (chloromethylisothiazolinone or CMT) and 2-methyl-4-isothiazolin-3-one (methylisothiazolinone or MT), which are the active ingredients in a 3:1 mixture (CMT:MT) sold commercially as Kathon. La méthylisothiazolinone, MT ou MI, est un puissant biocide et conservateur du groupe des isothiazolinones, utilisée dans les produits de soins et de nombreux autres produits (peintures notamment2) depuis 20053. C’est un produit allergène, source d’eczéma allergique chronique (y compris pour les produits rinçables, et avec de nombreuses rechutes4).
Suite aux scandales autour des parabènes, son usage s’est, en quelques années généralisé dans les produits cosmétiques et en 2010-2016 il est à l’origine d’ « une épidémie sans précédent d’allergie de contact », dont en Europe5, avec de possibles allergies croisées à l’octylisothiazolinone (OIT) et au benzisothiazolinone (BIT).
D’après des expériences sur le modèle animal (souris) les personnes sensibilisées au méthylisothiazolinone (MI) peuvent aussi réagir à l’OIT et au BIT si elles ont été exposées à une concentration suffisante de MI6.
Applications
La méthylisothiazolinone et les autres dérivés des biocides isothiazolinone sont utilisés pour contrôler la croissance microbienne dans des solutions contenant de l’eau7. Deux des biocides isothiazolinone les plus largement utilisés sont la 5-chloro-2-méthyl-4-isothiazolin-3-one (chlorométhylisothiazolinone ou CMT) et la 2-méthyl-4-isothiazolin-3-one (méthylisothiazolinone ou MT), qui sont les ingrédients actifs dans un mélange 3:1 (CMT:MT) vendu commercialement sous le nom de Kathon. Les produits Kathon sont vendus aux fabricants sous forme de solution concentrée contenant entre 10 et 15 % de CMT/MT.
Pour application, l’utilisation recommandée est entre 6 ppm et 75 ppm d’isothiazolones actives. Les applications biocides couvrent un spectre allant du stockage d’eau industriel aux unités de refroidissement, à des processus aussi variés que l’exploitation minière, la fabrication de papier, le travail des métaux fluides et la production d’énergie. De plus, une isothiazolinone, Sea-Nine 211 (4,5-dichloro-2-n-octyl-4-isothiazolino-3-one, DCOI), a rapidement remplacé le tributylétain comme agent antifouling dans la peinture des bateaux. Une étude récente a révélé la présence de DCOI à la fois dans les eaux portuaires et les échantillons de sédiments d’Osaka, au Japon, spécialement dans certaines zones8. Au niveau environnemental, les niveaux de DCOI considérés dans les marinas sont à présent considérés comme une question pour diverses espèces d’invertébrés marins (en)9. Les isothiazolinones sont aussi extrêmement toxiques pour les poissons10. La large utilisation d’isothiazolinones dans le domaine industriel a conduit à un très grand nombre de cas d’exposition humaine préoccupants, parfois dans des proportions épidémiques11. Cela survient d’abord, mais pas exclusivement, quand les travailleurs sont exposés à des solutions stockées au cours du processus de dilution, conduisant à des brûlures chimiques, des dermatites de contact, et des réactions allergiques4. Les expositions par inhalation sont aussi très courantes12.
Écotoxicologie
Alors que l’agriculture était autrefois et depuis des décennies la principale source depuis longtemps de biocide dans l’environnement, la lixiviation des peintures et enduits de façades des bâtiments2 et le rinçage (douches, bains, baignades) de cosmétiques et de produits de soins personnels et de nettoyage sont devenus de nouvelles sources préoccupantes de contamination de l’eau et de l’environnement13.
La Méthylisothiazolinone est depuis les années 2010 très présente dans les peintures2 et produits de soins. Les lixiviats et produits de rinçage en contenant sont de plus en plus retrouvés dans les eaux d’égouts et/ou de baignade, et comme plusieurs autres biocides elle se montre génotoxique pour certains animaux aquatiques (tests fait sur la truite)14.
Chez la souris une réaction inflammatoire/immunitaire apparait en trois jours et une sensibilisation est facile à induire6.
Un autre effet préoccupant, démontré par une publication de 2017 chez le planaire (5 espèces différentes utilisés dans ce cas comme espèce modèle) est que outre une perte d’intégrité épithéliale et de fonction neuromusculaire chez les planaires non lésés, cette molécule a inhibé la cicatrisation et de la régénération de planaires expérimentalement lésés15. Lors de cette étude des antioxydants se sont révélés être des antidotes des effets (chez le planaire). La même équipe avait précédemment montré que la Méthylisothiazolinone inhibait également la cicatrisation et la régénération de la queue de la jeune grenouille Xenopus laevis.
Une étude publiée en 2018 a apporté des données nouvelles concluant à une toxicité faible de ce produit pour la cellule (sur la base de tests in vitro sur des cultures de cellules de foie de truite arc-en-ciel (RTL-W1) et des cellules ovariennes de hamsters femelles (CHO-9), mais note que sur la base des données publiées, les valeurs de PNEC sont beaucoup plus élevées pour l’IM (0,5 μg/l) que pour la Terbutryne ou TB (0,003 μg/l) ou l’octhilinone ou OI (0,05 μg/l)13. Cette étude laisse penser que les risques environnementaux et écotoxicologiques posés par ce produit pourraient avoir été sous-estimés16.
Santé humaine
De nombreuses études ont montré que la MT 50 est allergénique (une exposition prolongée induit un risque plus élevée de dermatite allergique de contact17, avec parfois une réactivité croisée (avec la Pemphigoïde bulleuse pouvant conduire à l’hospitalisation18) et qui semble cytotoxique (toxique pour les cellules)19 et un travail récent (2017) a montré :
qu’il existe une grande différence de passage percutané (dans la peau) selon qu’il y a ou non rinçage du produit après qu’il a été posé sur la peau20,
que les estimations d’exposition utilisées pour des utilisations répétées de produits par jour étaient trop prudentes20.
En France, les résultats d’une étude du REVIDAL-GERDA (Réseau de Vigilance en Dermatologie Allergologie rattaché au Groupe d’Étude et de Recherche en Dermatologie Allergologie) ont montré qu’entre 2010 et 2012 la proportion de tests positifs à la méthylisothiazolinone a plus que triplé. Cette augmentation résulte probablement du remplacement de certains produits conservateurs (parabène, méthanal, méthyldibromoglutaronitrile (en)) par ce produit21.
À la suite des différentes publications scientifiques européennes, la MI a été déclarée « allergène de l’année 2013 » aux États-Unis et va être testée en routine dans la batterie d’allergènes standard nord-américaine22 et en 2013 la situation épidémiologique avait empiré.
DerMTe (ou dermatite) de contact allergique
Définitions
La derMTe de contact est une atteinte locale de la peau causée par le contact de celle-ci avec une substance étrangère. Il existe deux types de derMTe de contact : la derMTe de contact irritative et la derMTe de contact allergique.
La derMTe de contact irritative, plus fréquente, est causée par diverses substances irritantes provoquant une inflammation, une rougeur, une sensation de chaleur, une enflure et de la douleur.
Dans la derMTe de contact allergique, des cellules immunitaires spécifiques sont impliquées et sont sensibilisées à une substance. Ainsi, ces cellules vont s’activer à chaque fois que la peau entre en contact avec cette substance et causer une réaction cutanée, rougeur de la peau, la formation de vésicules, une desquamation et la formation de croûtes. À la différence de la derMTe irritative, la réaction peut apparaitre dans des régions du corps qui n’ont pas été en contact avec la substance allergène23.
Constat
La méthylisothiazolinone est couramment utilisée dans les produits, parfois en conjonction avec la méthylchloroisothiazolinone, avec dans les deux cas la possibilité d’induire une dermatite de contact allergique. La sensibilisation à cette famille de conservateurs a été observée dès les années 1980. En raison de l’utilisation accrue de conservateurs isothiazolinone depuis 2005, les cas signalés de contacts allergiques à ce produit ont été, eux aussi, en forte augmentation.
En 2013, la substance a été déclarée Allergène de contact de l’année 2013 (en), par l’American Contact Dermatitis Society24,25 et on observe un début d’épidémie en Europe5.
En 2015 une étude conduite dans huit pays européens (1er-31 octobre 2015) a montré que 6,0% des personnes testées (205/3434 patients) eu des réactions positive à ce produit (patch-test) et que la dermatite est localisée sur les mains dans 43,4% des cas puis sur le visage (32,7% de cas), les bras (14,6%) et les paupières (11,7%) mais 12,7% des patients ont eu une dermatite généralisée. La majorité des patients (79%) ont noté l’apparition de leur dermatite entre 2013 et 20155. Dans la plupart des cas, l’allergie a été attribuée à des cosmétiques, mais 7,3% des patients ont vu la réaction allergique apparaitre lorsqu’ils étaient dans des chambres nouvellement peintes et d’autres études ont montré que ce produit est aussi de plus en plus utilisé comme conservateur des peintures et enduits2 (sans être mentionné par l’étiquette souvent)5,26.
Principe de précaution
Le 13 décembre 2013, la société de cosmétiques Cosmetics Europe, après des discussions avec la Société européenne de la dermatite de contact (ESCD), a recommandé à ses membres de cesser l’utilisation de la méthylisothiazolinone (MT), pour les produits dermatologiques y compris les lingettes humides. Cette action est recommandée, dans l’intérêt de la sécurité des consommateurs, en ce qui concerne les effets indésirables sur la peau. Il est recommandé aux entreprises concernées, de ne pas attendre une intervention réglementaire, en vertu du règlement cosmétique27, mais de mettre en œuvre cette recommandation dès que possible28.
Avis de la Commission européenne
Le 27 mars 2014, le coMTé scientifique de la Commission européenne sur la sécurité des consommateurs (SCCS), a émis un avis sur la sécurité des méthylisothiazolinones. Ce rapport ne considère que la question de la sensibilisation de contact. Le coMTé a conclu :
les données cliniques actuelles indiquent qu’une concentration de 100 ppm (0,01 %), dans les produits cosmétiques, n’est pas sans danger pour le consommateur ;
pour les produits cosmétiques (y compris les lingettes humides), aucune concentration sûre de sécurité, pour l’induction de l’allergie de contact ou son déclenchement, n’a été démontrée ;
pour les produits cosmétiques, dits « à rincer », on considère qu’une concentration de 15 ppm (0,0015 %), est considérée comme sûre pour le consommateur, du point de vue de l’induction de l’allergie de contact. Cependant, aucune information n’est disponible quant au seuil de déclenchement27 et des études ultérieures montreront que les produits à rinçage sont presque aussi souvent en cause que sans rincage4.
Malgré des résultats d’études qui montrent tous sa responsabilité, en 2016 de nombreux produits cosmétiques (shampooing29, gel douche, après-shampooing, crème de douche, lait de toilette, lessive, lingette, etc.), contiennent ce conservateur décrié30,31.
Neurotoxicité démontrée avec le modèle animal dès 2002
Depuis le début des années 2000, des études conduites par des neuroscientifiques ont montré que la méthylisothiazolinone, même à faible dose32 est aussi « un biocide neurotoxique » et même « hautement toxique » pour les neurones en culture, bien que sans effets apparents pour les cellules gliales ; ce produit « perturbe l’association des tyrosine kinases de la famille SRC avec la kinase d’adhésion focale dans le développement des neurones corticaux »33. Il peut donc être à l’origine de troubles neurodégénératifs chez l’être humain.
En 2002 les résultats d’une étude34 ont clairement montré (in vitro) qu’il n’y a pas besoin d’une exposition chronique pour générer cette neurotoxité : une brève exposition suffit pour tuer les neurones mis en culture, via un processus toxique faisant intervenir le zinc et l’activation de la kinase régulée par le signal extracellulaire p44/42 (ERK) via une voie médiée par la 12-lipoxygénase34. Le processus de mort cellulaire implique aussi l’activation de la NADPH oxydase, la génération d’espèces réactives de l’oxygène, l’endommagement de l’ADN et la suractivation d’un enzyme : la Poly(ADP-ribose) polymérase, toutes se produisant en aval de la phosphorylation ERK34.
En 2012 on a montré in vivo chez les têtards de Xenopus laevis qu’une exposition chronique de neurones lors de leur développement à de faibles doses de MT 50 (doses largement sublétales) interfère avec l’excroissance normale des neurites, c’est-à-dire de tous les prolongement du corps cellulaire du neurone (axone ou dendrite)32. Ceci s’est traduite par des anomalies neurocomportementales de l’animal, sans mortalité accrue (en aquarium) ni de réduction de la capacité natatoire, et sans défaut anatomique externe apparent, mais avec des déficits de comportement d’évitement visuel qui coïncident avec des anomalies électrophysiologiques dans la fonction optique du Colliculus supérieur, et une susceptibilité accrue à des crises de type épileptiques)32. Les auteurs de cette études, tous comme ceux de l’étude précédente (publiée 10 ans plus tôt) ont invité à vérifier que d’autres biocides apparentés n’aient pas d’effets similaires sur les neurones, effets qui n’avaient pas été étudiés ni rapportés par la littérature scientifique avant 200232.
Excrétion et métabolites
Les analyses d’urines permettent de mettre en évidence un métabolite (acide N-methylmalonamique ou NMMA) de ce biocide et d’alors montrer qu’il y a eu passage percutané d’un produit en contenant (ou inhalation)35.

Les industries des détergents et des peintures utilisent depuis longtemps la MT 50 (méthylisothiazolinone) comme biocide/agent de conservation dans les formulations à base d’eau.
Jusqu’à présent, la législation exigeait que la MT 50 porte l’étiquette H317 (Peut causer une réaction allergique cutanée) si au-dessus de 1000 ppm, et la EUH208 si au-dessus de 100 ppm. Jusqu’à présent, cela n’a posé aucun problème pour ces formulations, car elles ont pour la plupart une concentration en MT 50 comprise entre 50 et 200 ppm.
Cependant, à partir de mai 2020, les liMTes de concentration de la MT 50 seront considérablement réduites à 15 ppm (H317) et 1,5 ppm (EUH208). En conséquence de cette modification réglementaire, ceux qui ne souhaitent pas modifier l’étiquetage de leurs produits devront chercher des substituts à la MT.
